It involves using a heated, saturated steam in an autoclave at high pressure. This sterilization method is commonly used in the medical and pharmaceutical industries to sterilize reusable equipment and glass ampules, vials, syringes, and plastic containers.
The specific temperature and duration of the sterilization cycle depend on the target of sterilization. Typically, steam sterilization cycles range from 1121&thinsp;&deg;C (30&thinsp;min) to 135&thinsp;&deg;C (10&thinsp;min) held at 2&thinsp;bar to achieve a 6-log reduction value. Other recommended operational parameters are 121&ndash;124&thinsp;&deg;C at 2&thinsp;bar for 15&thinsp;min, 126&ndash;129&thinsp;&deg;C at 2.5&thinsp;bar for 10&thinsp;min, or 134&ndash;138&thinsp;&deg;C at 3&thinsp;bar for 5&thinsp;min.. The operational conditions of the sterilization process are determined by factors such as the type of steam saturatedorunsaturated, temperature, pressure, and exposure time.

Figure&nbsp; illustrates a typical moist heat sterilization cycle and its three phases: evacuation, steam injection, and cool down. During the first phase, the sterilization chamber is evacuated to remove noncondensable gases. The second phase involves introducing steam to the sterilizer, increasing the temperature and pressure to around 135&thinsp;&deg;C and 3&thinsp;bar, respectively. These conditions are maintained for 5&thinsp;min before pressure release, evacuation, and cooling down. Temperature probes are used to monitor the sterilization process and ensure all materials in the autoclave reach the required temperature.
Moist heat sterilization works by denaturing and coagulating the proteins of microorganisms, resulting in their death. There are two types of autoclaves: gravity displacement sterilizers and high-speed prevacuum sterilizers. In gravity displacement sterilizers, steam is injected from the sides or top, and the lower density of steam causes it to flow to the top and push out colder air from the bottom. High-speed prevacuum sterilizers are equipped with vacuum pumps to remove air and decrease humidity before steam injection.
